1/ Super excited to share my first preprint from @hansen_lab! We used MINFLUX to track chromatin (H2B-Halo and Fbn2 locus) at an unprecedented 200 μs, then combined it with SPT to span μs-minutes (H2B) or SPT & Super-Res Live-Cell Imaging (SRLCI) to span μs-hours (Fbn2)

Out today in @NatureGenet, our group’s single-cell MPRA technology for testing activities of gene regulatory elements in whole tissues. Congrats to Siqi Zhao and the rest of the crew for their great work! nature.com/articles/s4158…
This is a really nice summary of the recent papers on E-P compatibility: doi.org/10.1016/j.molc…. Also nice to see that their results support our model that interactions between CREs are strength-dependent rather than dependent on specific TFs! (from genome.cshlp.org/content/32/1/85)
Our paper on insulators is out on Biorxiv! We tested the activity of insulators across thousands of genomic locations to figure out where insulators function. Oh, and I successfully defended today :) biorxiv.org/content/10.110…

We have a new version of this preprint! There are 2 key changes - we now show that scMPRA works in mouse retina (!), and we finally implemented the second U6 transcript to detect unexpressed promoters. Siqi worked really hard on this so please check it out 🙂
