Important work from @LukeJelen. The opioid system is involved in ketamine's antidepressant effect - well powered study with insight into the mechanism. Esp for ketamine responders, unequivocal reduction in efficacy when pretreated with naltrexone
1/ Just out in @NatureMedicine : we show the opioid system modulates ketamine’s effects in major depressive disorder, with naltrexone pre‑treatment reducing acute glutamatergic activity and acute antidepressant effects. nature.com/articles/s4159…

I wrote a little essay for Asimov about mitochondrial uncouplers. They are one of an unusual class of drugs that work without a molecular target, rather, the mitochondria are the 'target.' I see uncouplers as an exception that hints at the vast design space of small molecules
French munitions workers during WW1 mysteriously began losing weight. One of their explosive chemicals, called DNP, caused it. This became one of the first effective weight-loss drugs. But a newly-established FDA banned it for safety reasons in the late 1930s. By @Atelfo 🔻
Is methamphetamine meaningfully serotonergic or not? It seems like it can successfully blind MDMA while amphetamine cannot
Lot of interesting stuff in here, including 30% adjunctive IV methylphenidate, thorazine abortion @ 3-4 hours, and high rates of flashbacks (9%)

I strongly suspect psilocybin for PTSD will be more effective than MDMA for PTSD and psilocybin for depression
You think you’re pretty up-to-date on what’s going on in the psychedelic field, and then you see a study like this published 😂🐶 “Evaluating the Potential of Microdosing 1cp-LSD for the Treatment of Canine Anxiety: A One-Month Case Study” onlinelibrary.wiley.com/doi/10.1002/vm…

🧵THREAD: Our new study published in #lancetpsych! We found that pramipexole augmentation substantially reduced depressive symptoms in patients with treatment-resistant depression over 48 weeks of treatment. This is an important advance in how we treat depression! 1/6
We are learning so much from our clinical trial (PI @sdpnayak) examining Psilocybin Therapy for PTSD. Here are a few of the areas we're learning about: -How do SSRIs affect the intensity of experience? -What role can trauma-focused psychotherapy play following psilocybin?

This book is an amazing culmination of years of work and original thought and I can't really recommend more highly -- really pertinent for anyone interested in religion, psychedelics or mental health
Today is publication day for my book, Shamanism: The Timeless Religion. To celebrate, here are some of my striking clips of shamanic rituals that I came across while working on the book:
A lot of bad news these days, so I thought I’d share some good news. Last week, in a celebration of 150 people coming from all over the world to Buenos Aires, I married a revolutionary. Also, I got promoted to Assistant Professor. And yes, there were robots.
oh, this is extremely cool. post claims the reason point estimates of project completion times are frequently large underestimates is that they're estimates of the median of a lognormal distribution, which is heavy-tailed, and the mean is 1.6x the median
"That means the developer’s estimate, while far from perfect, is also far from meaningless: it’s the scale parameter of some distribution." When I read that earlier, my brain just fucking exploded. I'm still processing the implications. Link to the essay in first reply. I want…

People often claim that psychedelics have been used globally, for millennia, & in contexts of psychological healing. In today's @Guardian Long Read, I show what these stories get wrong & why the reality is far more interesting. Excerpted from Shamanism: The Timeless Religion
Honored that SHAMANISM has been named one of the best books of the month by @TIME! "Blending memoir, investigative journalism, and anthropological fieldwork, Shamanism is a deep dive into a religious tradition that is as mysterious as it is timeless." ~3 weeks until it's out!
