We found that human cells lacking JIP4 accumulate massive lysosomes that become filled with cystine, a disulfide form of the amino acid cysteine. This mirrors what happens when cystinosin (CTNS, the lysosomal cystine exporter) is knocked out

JIP4 deficiency causes a novel lysosome storage disease arising from impaired cystine efflux biorxiv.org/content/10.110… #biorxiv_cellbio

Glad to report our study on the "dual regulation of SLC25A39 by AFG3L2 and iron controls mitochondrial glutathione homeostasis" is out in @MolecularCell. x.com/MolecularCell/…
We first identified that human A39 contains a unique loop 1 with 4 matrix Cys. Through protein-mRNA correlation and CHX chase, we reasoned that A39 is dual regulated by two mechanisms, fast degradation (independent of Cys) and metabolite sensing (dependent on Cys). (2/n)
This feels like an early Christmas present. An incredibly robust picture and reference set for subcellular localization. My favorite proteins all make sense and there is so much to discover. Explore with the companion website and uncover new biology: organelles.czbiohub.org
So stoked to be sharing this! New proteomics map of human subcellular architecture. CRISPR + organelle IP mass spec at scale. Check out pre-print biorxiv.org/content/10.110… and organelles.czbiohub.org. 🙌 to fantastic team @czbiohub SF & collabs! 1/2
